LOC_ABRAMSON-Abramson Building Req ID: 43001

Shift: Days

Employment Status: Regular - Full Time

Job Summary

The research technician position is in the laboratory of Dr. Stella Chou, a Principal Investigator in the Hematology Research Division. Her research focuses on pediatric blood diseases, mechanisms of normal and pathologic hematopoietic development, and the use of pluripotent stem cells to model and create tools to improve diagnosis and management of blood disorders.

Research foci include:
1. Understanding how transcription factor GATA1 controls normal erythro-megakaryocytic development and how GATA1 mutations cooperate with genes on chromosome 21 to cause transient myeloproliferative disease and acute megakaryocytic leukemia in patients with Down syndrome.
2. Disruption of candidate chromosome 21 genes using CRISPR/Cas9 technology in pluripotent stem cells to examine their role in the blood abnormalities and predisposition to leukemia associated with Down syndrome.
3. Creation of customized pluripotent stem cells with rare blood cell phenotypes to generate red cell reagents that would improve antibody identification in transfused patients with sickle cell disease.
4. Identifying novel methods to expand pluripotent stem cell derived hematopoietic progenitors.
5. Adaption of next generation sequencing technology to improve red cell matching and prevent alloimmunization in patients with sickle cell disease.

Research technician duties include basic molecular biology procedures including DNA, RNA, and protein extraction; PCR, RT-PCR, and western blotting, as well as tissue culture including culture of cell lines, primary human blood cells, mouse and human embryonic stem cells, and human induced pluripotent stem cells. The research technician will also assist in general laboratory duties including maintenance of the laboratory, autoclaving, and ordering supplies.
